World number 32 Wilson’s break of 73 in the deciding frame at Meadowbank Sports Center in Edinburgh pitted him against either Hussein Favaei or Yan Bingtao in the round of 16.
O’Sullivan lost in a Triple Crown event for the first time in his career earlier this month, losing 6-0 to China’s Ding Junhui in the quarter-finals of the UK Championship.
Wilson also made scores of 73, 83 and 82 to secure his first career victory against world number one O’Sullivan.
Mark Selby set up a last-16 match with either Jack Lisowski or Ali Carter after beating Robert Milkins 4-1, while Zhou Yuelong and Mark Williams advanced after beating Barry Hawkins and Jimmy Robertson 4-0.
Judd Trump, who lost for the eighth time in his 147 career career in a 4-0 win over Mitchell Mann in the second round, defeated Jimmy Clark 4-1 and world number four Neil Robertson beat Joe Perry by the same score.
China’s Ding, who lost 10-7 to Mark Allen in the UK Championship final, lost 4-2 to Joe O’Connor and, in the all-Scottish competition, Anthony McGill beat Scott Donaldson 4-3.
And earlier on Wednesday, UK champions Allen sealed their place in the third round with 132 not out in a 4-2 win over Martin Gould.
Yan Bingtao took the deciding frame to defeat Liam Highfield 4-3, Ryan Day beat Liu Haotian 4-2, Lisowski dropped Alan Taylor, John Higgins beat Cao Yuping 4-3.
